Job Description More than 1’000 drugs have been associated with idiosyncratic heptotoxicity. Drug-Induced Liver Injury (DILI) therefore represents a major challenge for industry and regulatory authorities. Unfortunately, a drug’s idiosyncratic hepatotoxicity potential can only be recognized post-marketing, and DILI has therefore also been the most frequent single reason for withdrawing drugs from the market.
Your responsibilities include, but are not limited to:
• In vitro evaluation of reversible and irreversible inhibition potential on metabolizing enzymes and drug transporters • Determination of various hepatotoxicity readouts (e.g. ROS, mitochondrial tox, cell death, GSH adduct formation, GSH/GSSH ratio) • Further evaluation and development of the so-called Extended Clearance Model (ECM) in order to illustrate its ability to accurately anticipate and predict hepatocellular cell stress elicit by drug candidates • Establishment of correlations between process inhibition, intracellular bile acid accumulation and potential hepatotoxicity readouts (e.g. apoptosis, cell death, canalicular efflux pump upregulation) • Development of an integral DILI assessment decision tree (e.g. by introducing discovery of Adverse Outcome Pathways (AOPs))
Minimum requirementsWhat you’ll bring to the role: • Degree in organic/analytical/biochemistry or applied data science (MSc) • Willingness to engage in a University PhD program • High level of familiarity with data mining and big data concepts • Good attention to detail, an analytical investigative and questioning nature, with an innovative solution-oriented approach • A sharp and objective intellect, experienced in balancing big picture thinking with an investigative nature, drilling into detail as appropriate • Well-developed interpersonal skills with good presence; experienced presenter and communicator • A self-starter and decision taker; open, confident and persuasive, succeeding through influence • Good planning, prioritization, problem solving and organizational skills • Resilient, energetic and enthusiastic; responding constructively to challenging new ideas and inputs • Team player, ability to be flexible and adapt to a changing environment • Good knowledge of English language • Good IT skills particularly Word, Excel and PowerPoint
