Are you excited about proteases and their roles in health and disease? Are you intrigued by the vast design space of peptides and their integration into novel micro-and nanoagents for protease detection in vivo? If yes, the Responsive Biomedical Systems Lab (RBSL) at the Department of Health Sciences and Technology (D-HEST) of ETH Zürich might have the perfect job opening for you!

The RBSL develops diagnostic and therapeutic systems at the nano-and microscale with the aim to tackle a range of challenging problems in health care. For this purpose, strategies derived from chemical synthesis, synthetic biology, and nanofabrication techniques are applied. This interdisciplinary work is based on the convergence of several fields and integrates methods and ideas from chemists, physicists, computer scientists, engineers, and mathematicians.

Job description

Proteases are an exceptionally important and informative class of enzymes, making up 10% of all current drug targets and offering potentially useful diagnostic information. Developing robust methods for site selective detection of protease activity has the potential to advance understanding of fundamental disease mechanisms, aid in developing targeted therapies, and inform treatment. We recently developed a method to detect proteolytic activity via proteases responsive ultrasound contrast agents based on lipid shelled microbubbles (, patent submitted). Integration of suitable peptides for effective cleave by target proteases is key for clinical translation and the success of this methods.

The candidate will work in close collaboration with engineers, biologists, and clinicians to support the development of two kinds of agents for monitoring proteases. Concretely, the candidate will perform a comprehensive proteomic study of patient samples to identify protease profiles and corresponding peptide patterns to inform the design of application-tailored protease-responsive devices. He or she will also work with data scientists for sample analysis and determination of most suited peptides for integration as substrates, As one concrete clinical application, the project aims to lay the foundation for real-time in vivo assessment of inflammation in patients suffering from rheumatoid arthritis.

The workplace will be in Zürich. The position has a 100% pensum and is initially limited for 1 year with the possibility of prolongation.

Your profile

The ideal candidate for this project should hold a PhD in biochemistry or molecular biology with demonstrated expertise in mass spectrometry-based proteomics. Candidates with a biomedical or interdisciplinary background and solid biochemistry/molecular biology knowledge are also encouraged to apply. A strong candidate should ideally have prior experience with peptides, peptide chemistry, protein LC-MS and analysis of proteomic data. Some previous experience with enzymology, particularly proteolytic enzymes and detection of protease activity (e.g. with peptide substrates), especially in biological samples would be a strong asset.
